So, I have a level 20 ranger who right now is just using open aiming, no s/h skill. Fully doubled in Perception currently and has enough points to almost fully double in Ambush but has none right now. If I add the ambush, will he see a significant increase in his shots hitting the mark or does Ambush only help aiming if your hiding?

I believe perception is the only skill-based determinant of archery aiming. If ambush were a significant factor then those classes that could not train in it would have a really awful time aiming (wizards/empaths).
Someone correct me if there's literature to the contrary though, always up for learning something new!

Ambush helps with aiming while hidden. "Aiming from the open relies solely on the Perception skill, while aiming from hiding relies on the Perception and Ambush skills."
It also factors into your ranged AS after 40 ranks. Actually, more like 44, since the formula is (according to Nuadjha's awesome guide):
Ranged AS: DEX Bonus + Bow/Arrow Enchant (+50 max) + Ranged Skill + ((Ambush ranks – 40/4) + Perception ranks – 40/4)) + Spell effects (211, 215, Warcry, 1606, 425, etc.)
If you have read 'The Art of the Bow' on krakii, you should. It's very helpful.

Eh should have also asked: if you are 2x in ranged and 1x in brawl with a ranger, for example, do you get the ranged DS and the brawling parry DS? I'm mulling over going from short bow sniper to open aiming crossbow wielder on my forest gnome ranger. Using a bow as a forest gnome sucks because of the asinine RT mechanics for bows.

No you don't get brawl DS, you get bow DS while you have a bow in your left hand. That said archery RT is fine if you aren't playing the weakest race in the game.
